Wolverine Blend: structure and analysis

What it is

A two-compound blend supplied in a single vial: BPC-157 at 10 mg and TB-500 at 10 mg. The notation 10/10mg states each component's mass.

BPC-157 is a 15-residue synthetic peptide corresponding to a fragment of a protein found in gastric juice. TB-500 is the full 43-residue thymosin beta-4 sequence.

It is the simplest blend in the catalogue: two peptides, equal masses, no metal complex and no non-peptide components.

How it is produced

Supplied as a blend: the named components are combined and co-lyophilized into a single vial, with the mass of each component stated. The components are produced separately and characterised before combination.

This changes the analytical question. For a single compound, purity asks what share of the detected material was the target. For a blend there are several targets, and what matters is whether each named component is present at its stated mass — per-component identification and quantitation rather than one purity percentage.

Confirming identity

Two independent mass spectrometry confirmations. Fifteen residues against forty-three is a large enough difference that the two components are unambiguously distinguishable.

For TB-500 the identity result also has to confirm the full-length sequence rather than the short fragment sold under the same name, which is the substantive identity question in this vial.

Analytical considerations

Two peptides differing threefold in length separate well chromatographically, so resolving the components from each other is straightforward. Resolving each from its own deletion products is the real work, and for a 43-residue peptide that is demanding.

Quantitation of each component at its stated 10 mg is the result that matters, rather than a single purity percentage.

Neither component contains methionine, cysteine or tryptophan, so the oxidation pathways that affect blends containing Semax or MOTS-c are not the primary concern here.

What its certificate reports

A certificate for a lot of this material names the lot, names the issuing laboratory, and states for each parameter both the method used and the specification the result was measured against. Identity and purity are the two core results; net peptide content states how much of the fill weight is actually peptide.

A certificate belongs to a lot, not to a product line. When a new lot is released it is tested again, and the certificate published against it is that lot's certificate — a previous lot's result is never carried forward.
