What it is

BPC-157 is a synthetic pentadecapeptide — a chain of fifteen amino acids — corresponding to a fragment of a protein found in gastric juice. The sequence is produced synthetically rather than extracted, so the material in the vial has no biological source.

At fifteen residues it sits in the middle of the size range this catalogue covers: substantially longer than a tripeptide such as KPV, considerably shorter than the 43-residue TB-500.

How it is produced

Assembled by solid-phase peptide synthesis: the chain is built one residue at a time on an insoluble resin support, then cleaved from the resin, deprotected and purified — most commonly by reversed-phase high-performance liquid chromatography. The purified material is lyophilized to a dry solid.

The impurity profile that appears on a certificate is a record of where that process was imperfect: deletion sequences from an incomplete coupling, truncated chains, and species retaining a protecting group.

Confirming identity

Mass spectrometry confirms the fifteen-residue chain assembled as intended. At this length the mass difference produced by a single missing residue is unambiguous, so a deletion sequence is straightforward to detect.

The sequence contains no cysteine, so there is no disulfide bond to form or mis-form and no associated identity ambiguity.

Analytical considerations

Fifteen residues means fifteen coupling steps, and coupling efficiency compounds: the longer the chain, the more likely a deletion or truncation product appears in the chromatogram. This is the principal impurity class to expect and the one reversed-phase HPLC is well suited to resolve.

The absence of cysteine, methionine and tryptophan means the oxidation pathways that dominate for compounds such as Semax and MOTS-c are not the main concern here.

What its certificate reports

A certificate for a lot of this material names the lot, names the issuing laboratory, and states for each parameter both the method used and the specification the result was measured against. Identity and purity are the two core results; net peptide content states how much of the fill weight is actually peptide.

A certificate belongs to a lot, not to a product line. When a new lot is released it is tested again, and the certificate published against it is that lot's certificate — a previous lot's result is never carried forward.
